Pseudonocardia phyllosphaerae sp. nov., a Novel Actinomycete Isolated from Phylloplane of Rhizophora apiculata Blume
Abstract: A Gram-stain-positive actinomycete, designated strain 1LY6.1T, was isolated from the phylloplane of a mangrove plant (Rhizophora apiculata Blume). Phylogenetic analysis based on 16S rRNA gene sequences revealed that the strain was most closely related to Pseudonocardia nantongensis KLBMP 1282T (98.5%). The strain formed well-developed, branched substrate and aerial mycelium that fragmented into oval to rod-shaped and having smooth surfaces spores. The chemotaxonomic and morphological characteristics of strain 1LY6.1T were consistent with members of the genus Pseudonocardia, including meso-diaminopimelic acid as the diagnostic diamino acid. Major cellular fatty acids (> 10% of total fatty acids) were iso-C16:0 and summed feature 9 (10-methyl C16:0 and/or iso-C17:1 ω9c), and MK-8(H4) as the predominant menaquinone. However, the average nucleotide identity (ANI) and digital DNA-DNA hybridization (dDDH) values between the draft genome of this strain and its closely related type strain were 80.94 and 24.5%, respectively, which were below the proposed threshold values (95–96% for ANI and 70% for dDDH) used for species discrimination. Based on chemotaxonomic, phylogenetic, phenotypic, and genomic evidence obtained, we describe a novel Pseudonocardia species, for which the name Pseudonocardia phyllosphaerae sp. nov. is proposed. The type strain is 1LY6.1T (= TBRC 19001T = NBRC 116933T).
